Thin Layer Chromatography

1) Prepare solvent system (~100 mls) and let equilibrate in chamber containing Whatman #3 filter paper for 2 hours.

2) Draw a line on TLC plate ~2 cm up from the bottom and another ~1 cm down from the top.

3) Apply samples on 2 cm line, equidistant from each other - make sure sample will remain above the solvent level in the chamber.

4) Run plate until solvent front reaches the 1 cm line.

5) Remove plate from chamber and let dry in the hood.

6) When completely dry, place plate in Iodine chamber until yellow spots develop.

7) Circle spots immediately upon removing plates from chamber.

8) Spot corners of plate with radioactive ink (for orientation) and let ink completely dry.

9) Spray plate with Enhance (if needed).

10) Wrap plate in Saran Wrap and place in cassette with film (and intensifying screen if needed).
